MATERIALS AND METHODS
Syngeneic, orthotopic IT was undertaken in the Lewis rat using standard techniques. All grafts were procured, ffushed with lactated Ringer’s solution, and stored for 6 hours at 4 C prior to transplantation. Treated grafts also received rPSGL-Ig (Genetics Institute, Cambridge, Mass) 0.4 mg/kg intraarterial prior to cold storage and intravenously prior to reperfusion. Separate groups were used for survival and analysis. Tissue was collected from the terminal ileum at 4, 12, and 24 hours after IR and stored for analysis (n 4 per group and time point). Formalin-preserved, hematoxylin and eosin-stained samples were blindly graded by a single pathologist using the predetermined scale of injury: normal (0), superficial epithelial (1), full epithelial or lamina propria (2), submucosal (3), muscularis propria (4), full thickness (5). Competitive template polymerase chain reaction (PCR) was undertaken on frozen (70 C) samples using appropriate primers for interleukin 1 beta (IL1), IL6, and interferon-gamma (IFN). Noncompetitive PCR was undertaken for CINC/IL8. The myeloperoxidase assay (MPO) was undertaken as an indicator of PMN infiltration as previously described. 2
